A novel sodium benzoate analog (CH-1) decreases blood ammonia levels in experimental hyperammonemia

چکیده
Sodium benzoate has been used for the treatment of congenital hyperammonemic syndromes and hepatic encephalopathy for its ammonia lowering effect. A zinc-derivative of sodium benzoate (CH-1) with similar effects, is presented herein. We administered CH-1 at doses of 250, 500 and 1000 mg/kg body weight during 7 days to male Wistar rats after a portocaval shunt operation was performed as a model for experimental hyperammonemia. There was a decrease in blood ammonia levels in all groups receiving CH-1. A linear relationship (r= −0.487, PB0.01) was observed between the dose of CH-1 and blood ammonia levels. The effective dose 50 (ED50) was 320 mg/kg body weight (95% C.I. 176–581 mg) with a slope of 1.538 (Litchfield and Wilcoxon method). These results suggest that benzoic acid salts such as CH-1 may prove a new alternative for the treatment of hyperammonemia. Potential advantages of non-sodium salts of benzoic acid are discussed. © 1999 Elsevier Science Ireland Ltd. All rights reserved.

Late-Onset Ornithine Carbamoyltransferase Deficiency Accompanying Acute Pancreatitis and Hyperammonemia
Hyperammonemia related to urea cycle disorders is a rare cause of potentially fatal encephalopathy that is encountered in intensive care units (ICUs). Left undiagnosed, this condition may manifest irreversible neuronal damage. However, timely diagnosis and treatment initiation can be facilitated simply by increased awareness of the ICU staff. Here, we describe a patient with acute severe pancre...
متن کاملSuccessful use of alternate waste nitrogen agents and hemodialysis in a patient with hyperammonemic coma after heart-lung transplantation.
BACKGROUND Lethal hyperammonemic coma has been reported in 2 adults after lung transplantation. It was associated with a massive elevation of brain glutamine levels, while plasma glutamine levels were normal or only slightly elevated. In liver tissue, glutamine synthetase activity was markedly reduced, and the histologic findings resembled those of Reye syndrome. The adequacy of therapy commonl...
متن کاملFatal Hyperammonemic Brain Injury from Valproic Acid Exposure
BACKGROUND Hyperammonemia is known to cause neuronal injury, and can result from valproic acid exposure. Prompt reduction of elevated ammonia levels may prevent permanent neurological injury. We report a case of fatal hyperammonemic brain injury in a woman exposed to valproic acid. CASE A 38-year-old woman with schizoaffective disorder and recent increase in valproic acid dosage presented wit...

A 12-year-old female patient presented with nonspecific symptoms including fatigue, fever, and headache persisting for approximately 4 months. Plasma ammonia was 230 μg/dL (135 μmol/L) (critical value >187 μg/dL or 110 μmol/L). The patient was referred to a pediatric emergency center for urgent evaluation of hyperammonemia. Plasma and urine amino acid testing was performed to investigate the po...
متن کاملHyperammonemia in a Patient with Late-Onset Ornithine Carbamoyltransferase Deficiency
Ornithine carbamoyltransferase (OTC) deficiency is a urea cycle disorder that causes the accumulation of ammonia, which can lead to encephalopathy. Adults presenting with hyperammonemia who are subsequently diagnosed with urea cycle disorders are rare. Herein, we report a case of a late-onset OTC deficient patient who was successfully treated with arginine, benzoate and hemodialysis. A 59-yr-ol...
